Description: Ler was transformed with the pCLO461 construct (wild type Ac in GUS); the Ac element was cloned into a binary vector containing a 35S-GUS fusion; Ac was excised from the plasmid pJJ4361 on a Sal1-Sst1 fragment and ligated into the Sal1-Xho1 site of pSLJ732 which contains the 35S-GUS fusion; these transformed lines have been used to study cell patterning and clonal relationships in the roots of Arabidopsis, by following the pattern of GUS expression as a result of Ac excision (Scheres et al, 1994; Dolan et al, 1994).
